Uptake of P32 in resting cells of Clostridium perfringens.

Previous studies on resting cell metabolism of microorganisms relating to p32 incorporation into the intact cell have been carried out by Hotchkiss (1956) for micrococci, lactobacilli, and streptococci, and by Juni et al. (1948) for yeast. These studies in the main were concerned with turnover of various intracellular phosphoruscontaining compounds during conditions under which the organism was making protein from amino acids (Hotchkiss, 1956) or during ammonia assimilation (Juni et al., 1948). In an attempt to determine whether anaerobes could promote oxidative phosphorylation reactions, we initiated a study of the factors influencing the uptake of P3 by clostridia. Experiments reported here will show that during aerobic incubation of resting cells of this anaerobe, significant P32 incorporation into the intact cell occurred only in the presence of oxidizable substrate, and that this substratedependent P32 incorporation was inhibited by gramicidin and 2,4-dinitrophenol when ethanol was the substrate. These agents did not affect the oxygen uptake with ethanol. When glucose was the substrate, the p3 incorporation was influenced in a different manner in that DNP3 enhanced the incorporation and gramicidin
